When a booking is received does myallocator remove availability on all other channels?

Answer:

Yes, we call this feature Automatic Adjustment. When a booking comes up from a channel it updates myallocator, which in turn updates all configured channels.

You can activate this feature on the Automatic Adjustment Settings page in the Manage area.
